Lipid nanoparticles (LNPs) play an important role in therapeutic delivery, but their performance can depend on differences in composition, cargo loading, morphology, and stability that are difficult to resolve through bulk characterization alone. Understanding variation between individual particles can provide additional insight into formulation quality and help researchers evaluate how changes in lipid composition, cargo type, or processing conditions influence LNP populations.
In this webinar, brought to you by ONI, Colbie Chinowsky, and Claudia Zagami, will explore how super-resolution microscopy enables particle-level characterization of LNP formulations. Using commercially available LNP samples, they will examine differences in composition, cargo loading, ligand engineering, and batch heterogeneity, including changes associated with formulation stability and stress. The speakers will also discuss how single-particle imaging can complement established analytical approaches and support formulation development, optimization, and decision-making throughout drug development.
Topics to be covered:
- Identify particle-level heterogeneity within LNP populations that may be obscured by bulk analytical measurements.
- Compare LNP formulations and cargo loading, including differences between loaded and unloaded particles and different cargo or lipid compositions.
- Examine how super-resolution imaging can reveal formulation- and stability-related changes at the individual-particle level.
- Explore how particle-level characterization can complement established analytical methods during LNP development and optimization.
