Essential genes support cell viability and require precise, sustained expression. Many of these genes are regulated by CpG-island promoters that are densely occupied by transcription factors, leading to the prevailing model that multiple transcription factors cooperate to maintain gene expression. However, the individual contributions of these factors to chromatin accessibility and essential gene activation remain incompletely understood.
In this webinar, brought to you by EpiCypher, Ralph Grand will discuss research using rapid and reversible single and combinatorial transcription factor degradation in murine stem cells to dissect regulatory interactions between five key transcription factors. He will describe an unexpected regulatory architecture in which individual transcription factors autonomously drive chromatin opening and gene activation at largely distinct promoters. Grand will also demonstrate how single-molecule Fiber-seq enables direct visualization of transcription factor binding, nucleosome positioning, and associated changes in the chromatin landscape across kilobase-scale regions, providing new insight into essential gene regulation across development, homeostasis, and disease.
