The authors describe a novel molecular mechanism whereby downstream signaling in the mammalian target of the rapamycin (mTOR) pathway may proceed in a rapamycin-insensitive manner. This may help explain several recent reports of rapamycin-resistant effects resulting from cellular mTOR activation and also has implications for the design of drugs targeting the mTOR pathway.
