Embryonic stem (ES) cells can be derived and propagated in the presence of bone morphogenetic protein 4 (BMP4) and leukemia inhibitory factor (LIF) without serum or feeder cells. BMP4 induces expression of inhibitor of differentiation (Id) genes in ES cells via the Smad pathway, which, in conjunction with LIF/STAT3 pathway, is able to sustain self-renewal of stem cells while preventing their differentiation.
