Optimizing PacBio HiFi Long-Read Whole Genome Sequencing of Salivary DNA Using Oragene™ Collection Devices

Successful long-read sequencing starts with extracting high-quality DNA from saliva samples.

Brought to you byDNA Genotek Inc.

Obtaining high-quality, high-molecular-weight (HMW) DNA is key for optimal yield and performance of long-read sequencing with saliva samples. Salivary DNA stabilized in DNA Genotek™ Oragene™ devices, extracted using Nanobind™ kits, and sequenced on a single SMRT™ Cell on the Revio™ system typically generates greater than 20-fold HiFi coverage of the human genome, which is sufficient for comprehensive variant calling or de novo assembly.

Download this tech note to learn about the recommended Nanobind extraction workflow for saliva samples collected in Oragene™ devices, and the DNA QC metrics to achieve optimal HiFi sequencing performance.
