RNA-based therapeutics require scalable manufacturing strategies capable of producing high mRNA yields while efficiently using costly reagents. In vitro transcription (IVT) represents a critical step in mRNA production, but conventional batch approaches can present challenges as processes move toward larger manufacturing volumes. Fed-batch IVT, which gradually supplies reaction components during synthesis, provides an opportunity to improve reagent utilization, increase mRNA yield, and strengthen process control during scale-up.
In this webinar, brought to you by Beckman Coulter Life Sciences and Cytiva, Darius Menezes and Sebastian Blum will discuss the development and scale-up of a fed-batch IVT workflow for mRNA synthesis. They will examine how microfluidic feeding and real-time monitoring enabled rapid optimization of feeding strategies and feed stock concentrations in sub-milliliter reactions, resulting in mRNA yields of up to 15 g/L based on the initial IVT volume while improving nucleotide triphosphate (NTP) incorporation efficiency and reducing DNA template use. The speakers will also discuss the relationship between pH and crude mRNA concentration as a non-invasive process indicator and demonstrate how optimized conditions can translate from small-scale development to a closed, GMP-ready rocking bioreactor system.
Attendees will:
- Learn how fed-batch IVT strategies can increase mRNA yield while improving reagent utilization
- Explore how small-scale, microfluidic feeding approaches can accelerate optimization of feeding strategies and feed stock concentrations
- Discover how pH can serve as a non-invasive, real-time indicator of crude mRNA concentration during IVT
- Examine how optimized fed-batch IVT conditions can translate from sub-milliliter process development to GMP-ready manufacturing systems
