Optimizing adeno-associated virus (AAV) vector production means making critical decisions without capsid-loading data. Matrix noise in unpurified culture defeats single-particle detection, and established light-scattering and sedimentation assays demand high titers. These analytical limits push assembly checks downstream, often after the next production cycle has begun.
This application note provides a bench-scale cleanup protocol that moves capsid-ratio assessment to the bioreactor stage in three hours. Mass photometry resolves the constituent capsid populations from 1.5mL of prepared culture. Results match those from a full downstream purification run.
