HypoxyCOOL™, the new media conditioning system from Baker Ruskinn, accelerates the reduction of oxygen concentration in tissue/cell culture media to user-defined values by as much as two days. The system is the first commercialized solution for precisely controlling oxygen levels in liquid media used in stem cell expansion or any tissue culture process requiring accurate and stable oxygen levels.


“Exposing cells native to low-oxygen environments to normal atmospheric conditions causes abnormal cell reactions and reduces cell viability,” said Science Director Deborah Thibodeaux. “Even when gas-controlled incubators or workstations are used to control the atmosphere around cultures, they could still remain susceptible to oxidative stress due to higher concentrations of dissolved oxygen within cell growth media.”


HypoxyCOOLTM combats this oxidative stress by precisely reducing media oxygen concentration to user-defined values – in as little as three hours.

HypoxyCOOLTM solves these problems by enabling researchers to:


  • Enhance experimental control. Several variables that impact media conditioning are difficult to control using conventional processes. These variables include changes in media pH, potential microbial contamination, altitude, temperature and the presence of carbon dioxide. HypoxyCOOL helps control these variables and provides a repeatable conditioning protocol that improves the consistency of research results.
  • Increase productivity. Obtaining true, physiologically-relevant oxygen levels in media requires significant time – it may even take up to two days to achieve a dissolved oxygen concentration of 2% (see Figure 1). Moreover, this process must be repeated each time media conditioning is required. HypoxyCOOL accelerates the media conditioning process, allowing dissolved oxygen concentrations to reach 2% within as little as three hours. Additionally, oxygen levels stay low for up to 21 days, eliminating the need for reconditioning, significantly enhancing productivity in the lab.
  • Improve research results. In addition to the research conducted in Baker Ruskinn’s laboratory, independent research has shown that using in vivo-like oxygen levels in culture media stabilizes cell cultures, enhances gene expression and increases viability and transcriptional stability.
