Researchers who need to easily and accurately isolate a protein target from a crude sample frequently do so using immunoprecipitation (IP). The new Seize X Immunoprecipitation kits introduced by Pierce Chemical of Rockford, Ill., now offer improvements to this classic technique that enhance protein recovery and make SDS-PAGE analysis less confusing. These enhancements result in an IP method that offers better protein recovery without antibody contamination and allows the primary antibody to be conserved.

Traditionally, IP involves incubation of an antibody with cell lysate followed by precipitation of the antibody-antigen complex with immobilized protein A or G. The investigator then collects the support-bound complex by centrifugation, and elutes the bound proteins by boiling. Although this method is fairly reliable, it can be problematic because the purified proteins are contaminated with the antibody light and heavy chains that have been eluted along with the protein target. ...
