Acetylcholine, acting at receptors on endothelial cells, triggers the formation of nitric oxide, which diffuses to the adjacent smooth muscle cells to stimulate guanylyl cyclase, with the increased formation of cyclic GMP in turn eliciting muscle relaxation.
