Acetylcholine Transporter Antibody The new goat polyclonal antibody to vesicular acetylcholine transporter (VAChT) has been used for immunohistochemistry on rat and mouse tissue. It labels cholinergic cell bodies in the septum and nucleus basalis as well as cholinergic fibers in the brain stem and spinal cord. The antibody is claimed to be useful in Alzheimer's disease research because acetylcholine (Ach) is one of the first transmitters to be reduced in this disease. It also has applications for research on neuromuscular disease and the autonomic nervous system. CHEMICON International Inc. Temecula, CA The ChemiImager is designed to document, archive, and analyze gels, films, membranes, chemiluminescent samples, and plates. Using proprietary technology to thermoelectrically cool the charge-coupled device (CCD) camera, the system is claimed to minimize thermal noise in exposures ranging from 1/30 second to two hours. Real-time display optics have been engineered for white light applications as well as low light samples, such as chemiluminescence and fluorescence. The system includes reflective and transmitting white light illumination sources as well as an ultraviolet transilluminator. The Pentium-based processor incorporates AlphaEase software to provide image acquisition, enhancement, archiving, and analysis in a single package. Automatic 1D-multilane densitometry, 2D-spot densitometry, molecular weight determination, object distance measuring, and automatic colony counting reportedly can be accomplished in less than two minutes, while hard copies of images and analysis data can be output in less than 10 seconds. Weight readings are provided over a temperature range from 5¡C to 35¡C. Weighing units are displayed on the balances' 8-mm digital display. The balances contain a large, square plastic weighing pan to accommodate a variety of shapes and sizes. The pan is detachable for cleaning and maintenance. The front key panel is sealed for protection against dust and spills. The instruments include an indicator to monitor weighing stability, separate keys for power on/off and re-zero functions, and an automatic power-off feature to conserve battery life. They are available in 4-, 13-, and 25-mm sizes to handle volumes from 1 ml to 100 ml. A variety of membrane types are available to meet different application needs. These include Durapore (PVDF) for filtering aqueous solutions; PTFE for organic solvents; and HPLC-certified low extractable nylon and hydrophilic PTFE for aqueous/organic solutions. The company also offers syringe filters with tube outlets designed to eliminate air locks and spills when samples are filtered into small-volume vial inserts as well as robotic compatible filters for various workstations. Bedford, MA Tissue Dissociation Product Accutase is designed to maximize cell detachment and viability of a range of cell types, including fibroblasts, vascular endothelial cells, hepatocytes, CHO and BHK cells, and adherent tumor cell lines. It contains nonmammalian proteolytic and collagenolytic enzymes and reportedly has been shown effective in tissue dissociation of embryonic tissues for isolation of neurons and hepatocytes as well as in bone marrow cell isolation. The product is available in ready-to-use 100 ml 1x solution or in a 10x concentrate for tissue dissociation protocol. Cells are claimed to typically detach in five to 10 minutes and can be harvested and replated as usual. Protease inhibitors are not required. Innovative Cell Technologies Inc. La Jolla, CA The C4742-95 Progressive-Scan Cooled CCD Camera is designed for high-resolution and high-frame-rate measurements with sensitivity that is claimed to be comparable to that of conventional cooled CCD cameras. It incorporates a progressive-scan interline CCD that requires no mechanical shutter, thereby enabling quick measurement and high-speed electrical shutter function. The camera has 1,280 x 1,024 pixels with maximum 18Hz frame rate-2x2 binning and 9Hz for full resolution-at typically 10e read-out noise. Peltier cooling reportedly reduces dark noise and minimizes thermal drift. The camera is compatible with a variety of commercially available frame grabber boards by incorporation of AIA standard digital output. Hamamatsu Photonic Systems Bridgewater, NJ The Sure Blot CHEMI Hyb and Detection Kit is designed for chemiluminescent detection of digoxigenin-labeled probes following Southern blot, while the Fragile X CHEMI kit was developed for chemiluminescent Fragile X research. The reported benefits of the kits include longer probe shelf life, elimination of radioisotope hazards, comparable sensitivity to 32p, reduced exposure time, and the ability to strip and reprobe blots. The Sure Blot kit contains sufficient reagents for hybridization and chemiluminescent detection of 3,000 cm2 of membrane. The Fragile X CHEMI kit contains sufficient reagent to process 1,500 cm2 of membrane. It consists of a digoxigenin-labeled Fragile DNA probe and digoxigenin-labeled molecular weight markers. Oncor Inc. Gaithersburg, MD The Gemini Twin Shaking Waterbath is designed for running two procedures simultaneously in two independently controlled heated chambers. It features a low-profile chamber design to provide easy access to incubation containers for adding or removing reagents. In addition, removable stainless-steel chamber adapters hold vessels such as Tupperware containers, microplates, and trays in place during incubation without the need for lead weights. Chambers are heated to desired temperatures reportedly in less than 25 minutes, and the unit is said to be able to recover to the set temperature in less than 20 seconds after the cover is removed. A front-to-back reciprocating agitation motion is provided to both chambers and can be adjusted from 10 to 160 oscillations per minute. The instrument is useful for a variety of applications, including blot processing, cell culture plate incubations, and reverse hybridization assays. Robbins Scientific Corp. Sunnyvale, CA The Ultra-Lum 256-grayscale Thermal Printer produces publication-quality prints in about 4 seconds at less than 10 cents per image, according to the company. It offers a resolution of 8.9 dots per mm, 1.5 x and 2.0 x zoom capability, and portrait or landscape image orientation. The contrast and brightness dials can be set to control both the printer and the video display simultaneously to ensure that what is seen is exactly what is printed. The printer automatically adjusts to 115, 100, 220, or 240 voltage supplies. It complements the company's Modular Approach line of gel documentation systems, enabling users to print and digitally document color and fluorescent stains, autorads, blots, and ELISAs. Ultra-Lum Inc. Carson, CA The solenoid-operated Teflon Isolation Valves are designed for a variety of research applications involving high purity and aggressive fluids, including liquid and gas chromatography, protein purifiers, DNA and peptide synthesis, proportional selection, mixing, solvent selection, fraction collection, lab robotics, flushing, and stream switching. Wetted surfaces are completely isolated from the valve's metal solenoid parts. The valves are available in several configurations: two-way normally closed, two-way normally open, and three-way or mixing/selector with two, three, four, five, six, and eight inlets and one common outlet. Boonton, NJ The FluorImager 595 system reads a variety of fluorescent dyes for polymerase chain reaction (PCR) product labeling, such as FAM, TET, HEX, R6G, Bodipy, Cy3, and tetramethylrhomadine. It features a dual-line laser that offers two-color imaging of in-line standards and double-loaded gels for improved allele-calling accuracy and throughput. The system facilitates band recovery from SSCP gels as well as quantitative imaging of TaqMan microplates, chemifluorescent blots, and fluorescent protein gels. Fragment Analysis, ImageQuant, and Genetic Typing software programs for Windows NT workstations provide system control and capabilities for analysis of image data produced by the system. Molecular Dynamics Sunnyvale, CA The Mouse B Immunocolumns, marketed under the brand name "cellect," have been developed for the isolation of highly purified populations of murine B lymphocytes (B cells), which are the only antibody-producing cells and key cells responsible for immune response. The columns use an affinity chromatography isolation procedure based on negative selection, which reportedly gives higher yields of unaltered, fully functional cells than do similar techniques. Mouse T lymphocytes as well as monocytes and macrophages are removed by the immunocolumns. Because B cells are sensitive to activation, negative selection ensures that the recovered cells behave as they would normally. The company claims that the new columns can isolate 85 percent pure mouse B cells with specific yields of 50 percent. The recovered cells are fully functional, based on mitogen functional studies. Biotex Laboratories Inc. Edmonton, Alberta, Canada The Eclipse E800 microscope, featuring a CF160 infinity optical system, offers a magnification range from ultra-low total visual magnifications of 5 x up to 3,000 x. Users may capture images in film, digital, and video formats. The system is based around a longer objective parfocal length of 60 mm and a larger microscope objective barrel diameter of 25 mm. Combined with the optimum 200 mm focal length microscope tube length, this increased size reportedly results in longer working distances, higher numerical apertures, and the widest magnification range and documentation field sizes now available. More than 40 objectives are currently available for the E800 system. Nikon Inc. Melville, NY The MicroCell is a liquid cell that enables researchers to acquire scanning probe microscopy data in liquid environments. It can operate with a variety of microscopy systems, including contact and intermittent contact atomic force, lateral force, force modulation, and phase detection. The cell holds about 100 µl of liquid. Fluid input and output ports allow fluids to be rapidly exchanged without the need to remove the liquid cell from the instrument, which reportedly enables users to investigate responses to liquid in real time. The cell is useful in a variety of research applications in cell biology, molecular biology, plant biology, structural biology, drug discovery, and protein-ligand binding.
