However, TFIID resisted purification from higher organisms, preventing detailed studies of the process. We were interested in TFIID because our work implied it was a target for adenovirus E1A transactivation. The breakthrough in this area came in 1988 with reports from Leonard Guarente and Phillip A. Sharp (Massachusetts Institute of Technology) (S. Buratowski, et al., Nature, 334:37-42, 1988), and Pierre Chambon (Institute of Biological Chemistry, Strasbourg, France) (B. Cavallini, et al., Nature, 334:77-80, 1988) that a protein from yeast could replace human TFIID for in vitro transcription with all other TFs and RNA polymerase from human cells. Based ...
